IL-36γ promotes psoriasis-like features in keratinocytes in an imiquimod-induced murine model of psoriasis

Introduction Psoriasis is a recurrent, chronic inflammatory skin disease with complex pathogenesis. The imposes heavy burden on patients. Interleukin (IL)-36γ belongs to the IL-36 family and predominantly expressed by epithelial cells. IL-36γ upregulated in psoriasis lesions. However, effects of keratinocytes remain unclear. Material methods Eighteen IL-36γ-deficient mice were divided into three groups: vaseline group, imiquimod (IMQ) IMQ/IL-36γ group. Vaseline or IMQ was administered for 6 consecutive days. severity psoriasis-like lesions evaluated using modified Area Severity Index (PASI) scoring system. Production cytokines expression differentiation markers assessed immunohistochemistry. Results IMQ-induced significantly more severe IMQ/IL-36γ-treated compared vaseline-treated IMQ-treated mice, as shown an exacerbated phenotype, increased numbers blood vessels, infiltration cells, epidermal thickness. Expression loricrin keratin 5 decreased following treatment IL-36γ. Levels IL-17A, interferon-γ, β-catenin Dickkopf-related protein 1 elevated within psoriatic stimulation. Conclusions Together, these data showed that contribute abnormal proliferation keratinocyte-related proinflammatory cytokines, suggest may play important role pathogenesis psoriasis.
